Hydrolysis of bradykinin by stem bromelain.

چکیده
During the course of studies on the anti-edema and anti-inflammatory actions of stem bromelain, a potent kininase activity was observed (1). The present paper describes the analysis of the bonds of bradykinin which are susceptible to the action of stem bromelain. Minoh, Osaka) dissolved in 3 ml of 5 mM phosphate buffer, pH 7.5, containing 2 mM (3 mercaptoethanol, was mixed with 3 ml of purified stem bromelain (7.2 nag: 0.218 lomoles) preincubated in the same buffer at 37'C for 20 min for full activation. Twenty microliter aliquots of the reaction mixture incubated at 37 °C were withdrawn every 5 min for assay of rat uterus smooth muscle contracting activity. The contracting activity decreased to 0.5 of the original after 15 min incubation. The resultant reaction mixture was stored at-20°C until required. Enzyme solution without added bradykinin was treated in parallel as control. An aliquot of the reaction mixture was heated in a boiling water bath for 10 min to inactivate the enzyme and dinitrophenylated at pH 9.0 at 40'C in the dark for 2 hr (2). The DNP derivatives were separated into ether-soluble and aqueous fractions by extracting with

N-Benzoyl-L-serine Methyl Ester Catalysed by Bromelain and by Papain ANALYSIS OF MODIFIER MECHANISMS
1. N-Benzoyl-L-serine methyl ester was synthesized and evaluated as a substrate for bromelain (EC 3.4.22.4) and for papain (EC 3.4.22.2). 2. For the bromelain-catalysed hydrolysis at pH 7.0, plots of [SO]/vi (initial substrate concn./initial velocity) versus [SO] are markedly curved, concave downwards. 3. Analysis by lattice nomography of a modifier kinetic mechanism in which the modifier is su...
متن کاملActinopyga lecanora Hydrolysates as Natural Antibacterial Agents
Actinopyga lecanora, a type of sea cucumber commonly known as stone fish with relatively high protein content, was explored as raw material for bioactive peptides production. Six proteolytic enzymes, namely alcalase, papain, pepsin, trypsin, bromelain and flavourzyme were used to hydrolyze A. lecanora at different times and their respective degrees of hydrolysis (DH) were calculated. Subsequent...
متن کاملKidney therapeutic potential of peptides derived from the bromelain hydrolysis of green peas protein
Objective(s): Kidney disease is a global health problem that needs a solution to its therapy. In the previous study, we found that protein hydrolysate of green peas origin of Indonesia hydrolysed by bromelain (PHGPB) showed improve kidney function in cisplatin-induced nephropathy rats. In this study, we investigated the effect of PHGPB to obtain effective dose that exe...
متن کاملBioactivities of fish protein hydrolysates from defatted salmon backbones
Bioactivities of bulk fish protein hydrolysates (FPH) from defatted salmon backbones obtained with eight different commercial enzymes and their combinations were tested. All FPH showed antioxidative activity in vitro. DPPH scavenging activity increased, while iron chelating ability decreased with increasing time of hydrolysis. All FPH showed ACE inhibiting effect which depended on type of enzym...
متن کاملIsolation and characterization of two forms of an acidic bromelain stem proteinase.
Two forms of an acidic bromelain proteinase isolated from crude bromelain, an extract from pineapple stem, were found by a two-step FPLC purification procedure. The basic main components were removed by cation exchange chromatography and the breakthrough fraction was further resolved by anion exchange chromatography into 15 protein fractions, only two of which, called SBA/a and SBA/b, were prot...
